Supplemental Figure 1

(07-101907SuppFigure1.tif; 3.29 MB) CCL21 and NPY are found in the same sucrose fractions. Differentiated NG-108 cells were transfected with either CCL21-EGFP, NPY-mRFP or EGFP alone. After lysis, supernatants were centrifugated O/N in a linear sucrose gradient and eleven fractions for each conditions were processed for western blotting. CCL21-EGFP was detected in fractions 5, 6 and 7, which correspond to the same fractions where NPY-mRFP was detected. On the contrary, EGFP was observed mainly in the first three fractions.
Supplemental Movie 1

(07-101907SuppMovie1.avi; 3.93 MB) In transfected neurons CCL21-EGFP-filled vesicles originate in the golgi/trans-golgi network in the soma. From there, they are preferentially transported into the axon but can also be observed in the dendrites of the cells. Pictures of this movie have been taken with a sample rate of 3Hz. In total 150 s have been recorded and compressed to 30 s. Magnification is 200X.
