  Familial Alzheimer's disease mutations in C99-GFP do not alter ERC localization of the protein in DAPT-treated cells. Fluorescence microscope images showing that in control cells, expression levels of C99-GFP and the three different mutants (C99/EGg, C99/V717F, and C99/V717I) were below detection levels (e–h). Treatment of WT C99 expressing cells (i) and mutant C99 expressing cells (j-l) with DAPT resulted in the stabilization and increased levels of the proteins. Both WT and mutant C99 accumulated to the ERC in a similar manner after DAPT treatment (i–l, and magnifications in m–p; arrows indicate the ERC localization). a–d show phase contrast images of the control cells.
