Relative cell growth was significantly reduced in subclones stably knocking-down CCND1 (G18+shRNA-CCND1, G11+shRNA-CCND1) as well as in subclones stably complemented with the genomic BAC-clone insert of the CDKN2A/B gene (G18+BAC-CDKN2, G11+BAC-CDKN2), irrespective of the genomic site of integration. In contrast subclones stably modified with the selection cassette (G18, G11), with the empty pSUPERdL vector (G18+empty vector) or with an unspecific control shRNA (G18+shRNA-Luc) display the same doubling time characteristics typical for Granta-519 cells (G).
