Figure 2. Uniform VEGF distribution in ischemic muscle reveals a therapeutic window determined by microenvironmental concentrations. Comparison of the parental polyclonal VEGF population and the 100% clone shows marked differences in the characteristics of induced vessels and function, despite the fact that both populations express similar total amounts of VEGF in vitro and in vivo. Blood flow recovery and collateral growth are much more pronounced with the 100% clone, showing that microenvironmental concentrations are critical. Comparison between the 3 monoclonal populations shows that neither the 10% nor the 180% clone induce collateral growth or improve blood flow, revealing a previously unrecognized therapeutic window determined by microenvironmental VEGF concentrations.
