Figure 1. CYC10268 inhibits gene expression downstream of CSF-1R and CSF-1 immunomodulation in BMM. A) CSF-1-starved BMM were treated with CSF-1 and/or 5 µM CYC10268 over 24 h. Total RNA was collected at the indicated time points and reverse transcribed to cDNA. uPA, tlr9, and apolipoprotein E mRNA levels were estimated by real-time polymerase chain reaction (PCR) using SYBR green amplification and an ABI7000. Gene expression levels were normalized to hprt expression. B) BMM, pretreated with vehicle or 5 µM CYC10268, were primed with CSF-1 or IFN{gamma} for 20 h before treatment with 10 ng/ml LPS for 24 h. ELISAs were performed on supernatants for TNF{alpha}, IL-6, and IL-12. Data points represent the mean of triplicates, and SD are displayed. These data are representative of at least 3 independent experiments.
