Table 1. Transposition assay data from the use of chimeric transposases and modified target plasmids.

Experiment Total Donor plasmids recovereda Total number of transposition events Transposition frequencyb Fold Increasec

Control Mos helper + pGDV1 4.59 x 106 68 1.48 x 10-5
Gal4-Mos helper + pGDV1 2.08 x 106 48 2.32 x 10-5 1.6
Gal4-Mos helper + pGDV1-UAS 1.80 x 105 53 2.96 x 10-4 20 (12.7)
Gal4-piggyBac helper + pGDV1 6.12 x 105 12 1.96 x 10-5
Gal4-piggyBac helper + pGDV1-UAS 2.93 x 105 67 2.28 x 10-4 11.6

a The total number of donor plasmids recovered is estimated by the number of Amp resistant colonies recovered.

b The transposition frequency is calculated by dividing the number of confirmed transposition products recovered, by the total number of donor plasmids recovered. These data are the cumulative data from 3 independent injection and recovery experiments.

c The fold increases for the Mos1 experiments are relative to the Control Moshelper + pGDV1 experiment except for the number in parentheses, which represents the fold increase in transposition when using pGDV1-UAS target plasmid, compared to the standard pGDV1 target plasmid. The fold increase for the piggyBac experiment is based on the increase observed when using the pGDV1-UAS target plasmid compared to the standard pGDV1 target plasmid.
