Human atheromatous plaques stimulate thrombus formation by activating platelet glycoprotein VI

Supplemental Data

Human blood anticoagulated with hirudin was perfused at high wall shear rate (1500 s-1) over a plaque-coated surface that was fluorescently labeled with anti-collagen type III antibody. Flow is from left to right. The real-time video clip displays thrombus formation using bright field, fluorescence and reflection interference contrast microscopy (RICM). At t= 0, in comparison to the bright field images the RICM-technique shows plaque material close to the surface (grey area, distance approximately 10 nm) and more distant from the surface (white areas, 20-30 nm distance). The focal plane was changed in order to assess the height of the thrombus. (mov, 21 MB)
