(FigureS1.tif; 11.4 MB) NOD1/2 gene expression in non-CF and CF bronchial epithelial cells. The expression of NOD1/2 mRNA was determined by semi-quantitative RT-PCR of RNA derived from 16HBE14o- cells and CFBE41o- cells. GAPDH expression was also analyzed to ensure that equal amounts of RNA were used in each reaction. Results are representative of two independent experiments.

(FigureS2.tif; 7.0 MB) Effects of NOD1/2 ligands on NF-κB activation in non-CF and CF bronchial epithelial cells. 16HBE14o- and CFBE41o- cells were transiently transfected with the NF-κB-luc construct. The transfected cells were then stimulated with TLR2 agonist PGN (10 μg/ml), NOD2 agonist FK565 (10 μg/ml) and NOD1 agonist MDP (10 μg/ml) for 3.5 hr. Luciferase activity was assessed in agonists-treated and untreated cells. Results represent the mean ± SD from two independent experiments performed in triplicates.
